Enveda’s oral obesity pill shows strong GI safety in Phase 1, paving way for trials in patients off GLP-1 drugs

Aug 19, 2026·read at BioSpace ↗Positive readoutPositive

Enveda reported that its Phase 1 study of the oral obesity candidate ENV-308 demonstrated an exceptional gastrointestinal safety profile in healthy volunteers. The topline data, released on August 19, 2026, showed no serious adverse events related to the gut, a key concern for obesity therapies.

Current GLP‑1 receptor agonists such as Eli Lilly’s Zepbound and Novo Nordisk’s Wegovy drive significant weight loss but often cause nausea, vomiting, and other GI side effects that limit patient adherence. Moreover, many patients regain weight after stopping these injections, highlighting an unmet need for a tolerable, oral alternative.

ENV-308 is designed to mimic the hormone N‑lactoyl‑phenylalanine (Lac‑Phe), which rises after exercise and suppresses appetite in animal studies. Enveda engineered the molecule to remain active long enough for once‑daily oral dosing, addressing the rapid clearance of natural Lac‑Phe.

With FDA clearance already obtained for human testing, the company plans to move into a trial that enrolls individuals discontinuing GLP‑1 therapy, aiming to see whether the tablet can preserve weight‑loss benefits without the gastrointestinal drawbacks of injectable drugs.
