The phase II PEMDA-HN trial evaluated adding the STAT3 antisense oligonucleotide danvatirsen to pembrolizumab in patients with untreated recurrent or metastatic head and neck squamous cell carcinoma.
The study was stopped early after a second interim analysis because the combination did not increase the overall response rate compared with pembrolizumab alone.
Safety was considered manageable, but patients receiving the combination experienced a higher incidence of certain adverse events than those on pembrolizumab monotherapy.
