INBRX-106 plus pembrolizumab shows higher response rate than pembrolizumab alone in first-line PD-L1-positive head-and-neck cancer
The HexAgon-HN trial evaluated the OX40 agonist INBRX-106 combined with pembrolizumab in patients with treatment-naive, PD-L1‑positive metastatic or unresectable head‑and‑neck squamous cell carcinoma.
The combination produced a higher overall response rate compared with pembrolizumab monotherapy and was associated with greater proliferation and activation of CD8‑ and CD4‑positive T cells.
Safety was manageable, with no treatment‑related deaths and most adverse events being low grade. Progression‑free survival results and the start of the phase 3 portion of the study are expected in late 2026.
