Nivolumab added to standard radiation and hormone therapy yields 90% two-year disease control in high-risk prostate cancer

A Phase 2 trial evaluated the combination of nivolumab with standard radiation and hormone therapy in patients with high‑volume grade group 5 prostate cancer.

The study reported a 2‑year freedom from biochemical recurrence rate of 90.3%, with a confidence interval of 74.3% to 98.0%.

Safety findings showed grade 2 and grade 3 acute adverse events in 6.3% of patients each, and no grade 4 or higher events were observed.

Researchers identified the Ricketts Immunosuppression Score as a potential predictive biomarker; higher scores were linked to early pathological and radiographic responses, while lower scores were associated with a greater risk of metastatic progression (HR 9.256, p=0.044).
